- Amy Rose (voiced by Grey Griffin) - the game's protagonist. While not as fast as Sonic the Hedgehog, she possesses the Piko Piko Hammer as her weapon, which allows her to destroy most of the existing obstacles.
- Sally Acorn (voiced by Kath Soucie) - the Princess of Mobius and Amy's closest friend, possessing Ring-Blades as her weapon, allowing her to slice through metal and repel attacks from enemies.
- Nicole the Holo-Lynx (also voiced by Kath Soucie) - an artificial intelligence able to materialize herself, being Sally's sidekick.
- Cream the Rabbit (voiced by Tara Strong) - Amy's longtime sidekick.
